Deformation‐based morphometry analysis shows early degeneration in Nucleus basalis of Meynert in Alzheimer’s disease

چکیده

Background Nucleus basalis of Meynert (NbM) is the main source cholinergic projection to cerebral cortex, including entorhinal cortex (EC). Both cells NbM and their receptive targets in EC show degeneration during early stages Alzheimer’s disease (AD). While precise delineation region on T1w scans difficult due limited spatial resolution contrast, we sought quantify along trajectory confirm recent studies suggesting that happens before using deformation-based morphometry (DBM). Method Data included MRI 1473 subjects from Disease Neuroimaging Initiative (ADNI) dataset. Resolution was increased 0.5 mm isotropic voxel-size non-linear registration an ADNI-based unbiased symmetric template. The resulting deformation fields were used compute Jacobian determinant map for each subject as a proxy local volume. masks mapped our high-resolution template mean values inside these calculated groups with different baseline diagnosis, sex- age-corrected z-scores calculated. Two-sample t-tests then compare scores between groups. Result Figure 1 shows DBM score both regions Normal Controls (CN), early- late-stage mild cognitive impairment (EMCI LMCI) probable AD dementia. Results corrected multiple comparisons showed significant difference CN EMCI LMCI left right NbM. A also observed (p-value = 0.0024). Z-scored volumes did not groups; However, results differ significantly hemispheres. We expected differences group (P<0.001). Conclusion Our while macroscopic changes emerge MCI advance toward later stages, starts sooner, preliminary MCI. This points MRI-based measurements potential biomarkers detection marker burden.

Comparative analysis of the nucleus basalis of Meynert among primates.

Long projection axons from the Ch4 cell group of the nucleus basalis of Meynert (nbM) provide cholinergic innervation to the neurons of the cerebral cortex. This cortical cholinergic innervation has been implicated in behavioral and cognitive functions, including learning and memory. Recent evidence revealed differences among primate species in the pattern of cholinergic innervation specific to...

The nucleus basalis of Meynert in 20 definite cases of Creutzfeldt-Jakob disease.

The population of neurons and the neuronal size in the nucleus basalis of Meynert (nbM) were studied in 20 patients with definite Creutzfeldt-Jakob disease (CJD). When compared with a normal control group, the 20 CJD brains showed a significant loss of neurons and reduction of neuronal size, mainly in the middle level of the nbM and mostly affecting the right side. Since these findings show som...

Cell loss from the nucleus basalis of Meynert in Alzheimer's disease.

We examined the degree of neuronal loss from the nucleus basalis of Meynert (nbM) in two groups of Alzheimer patients differing in the degree of intellectual impairment. Significant cell loss from the nbM was found only in the more severely demented group of patients. Mean cell counts (per 10 mu paraffin section) were compiled separately for the anterior, intermediate and posterior subdivisions...

Yokukansan and Yokukansankachimpihange Ameliorate Aggressive Behaviors in Rats with Cholinergic Degeneration in the Nucleus Basalis of Meynert

Yokukansan (YKS) and yokukansankachimpihange (YKSCH) are traditional Japanese Kampo medicines. The latter comprises YKS along with the medicinal herbs Citrus unshiu peel and Pinellia tuber. Both of these Kampo medicines are indicated for the treatment of night crying and irritability in children and for neurosis and insomnia in adults. In recent clinical trials, YKS exhibited ameliorative effec...
